چکیده

Background: Matrigel (extracellular matrix) can improve the growth of many cell types in vitro. Objective: The aim of the present study was to determine the effect of Matrigel on the development of 2-4 cells human embryos in culture. Material and Methods: Surplus 2-4 cells human embryos, resulting from ICSI, were divided into two groups (control and test). Quality of embryos in both groups was morphologically similar. The test group (n=140) was cultured in Hams’ F10 supplemented with 10% human serum albumin and 150 μl liquid Matrigel. The control group (n=140) was cultured in the same medium devoid of Matrigel. Embryos were cultured for an additional 4 days and their morphology was assessed every 24 hours. Both groups were then statistically compared. Results: The percentage of the human embryos that reached the morula stage in the control and test groups were 79.2% and 80%, respectively (p>0.05). However, 36.4% of embryos reached the blastocyst stage in the test group as compared to 5.7% in the control group after 144 hours in culture. This difference was statically significant (p <0.01). In addition, culture of embryos on Matrigel and medium versus medium alone significantly improved in vitro hatching (25.7% versus 3.5%; p <0.01). Conclusion: Matrigel at low concentration enhances human blastocyst formation and hatching in vitro.

Matrigel Enhances in vitro Bone Differentiation of Human Marrow-derived Mesenchymal Stem Cells

Objective(s) The use of co-culture cells as well as extra cellular matrix are among those strategies that have been employed to direct mesenchymal stem cell (MSC) bone differentiation in culture. In this regard, there is no study considering the effects of Matrigel on mesenchymal stem cell (MSC) in vitro bone differentiation. This was the subject of the present study. Materials and Methods ...

P-126: Stem Cell Factor Increases Blastocyst Formation in Mouse Two-Cell Embryo Culture

Background: It is demonstrated that c-Kit( receptor of stem cell factor) mRNA is expressed in late 2-cell stage to the expanded and hatched blastocyst and the stem cell factor (SCF) transcript is detected in the oviduct and uterus. The aim of this study was to investigate the effect of different doses of SCF on mouse 2-cell embryo development in vitro. Materials and Methods: 4-6 weeks old femal...

P-63: Comparison of The Efficacy of Two Culture Media (Handmade vs. Commercial) on In Vitro Development of Vitrified and Non-Vitrified Mouse Embryos

Background: While assisted reproductive techniques (ARTs) are considered to be safe, many concerns exist about the safety of these techniques. Two important and indispensable parts of these techniques are in vitro culture and vitrification. While much progress has been made in developing culture conditions, it remains suboptimal. In order to investigate the effect of culture medium on in vitro ...

Heparin-binding epidermal growth factor significantly improves human blastocyst development and hatching in serum-free medium.

The purpose of this study was to determine the effect of heparin-binding epidermal growth factor (HB-EGF) on human embryo development in vitro from days 2 to 14 post-insemination. Embryos were cultured in a complex serum-free medium (CSFM3) in the absence and presence of 1 nM and 100 nM HB-EGF. Development to the blastocyst stage of A-C-grade embryos (A grade = highest quality) was improved in ...

Effects of blastocyst artificial collapse prior to vitrification on hatching and ‎survival rates and the expression of klf4 gene in mouse embryos

Although the rate of blastocysts implantation of embryos is higher than previous stages but their survival rate is lower than them, which could be attributed to the completely filled blastocoel cavity with liquid and increased possibility of the formation of ice crystals. This liquid could prevent the penetration of cryoprotecting materials into the embryos. In this study, we reduced the volume...
